Dearest listers,
We are interested in buying an hardware autofocus upgrade for our HCS microscope (the CRISP system, namely). We have it here for testing and we would like to be able to do multi-well imaging. This would imply that, when switching wells, one could turn off and back on the autofocus.
Does anyone know if this is possible through the GUI? Or through a bean shell script? We're novices when it comes to programming plugins, but we'd be willing to create and share one for the purpose later; for now we just want to test the hardware.
Thank you! And a happy new year! Best, Pedro Almada Research and Microscopy Technician Unidade de Imagiologia Celular,

Not exactly what you want, but if you check the Autofocus checkbox in the MDA window (and select the CRISP), Micro-Manager will engage the Autofocus before each position/timepoint and then disengage the autofocus. As long as the difference in height between positions is not too large, this should work for you. B.t.w., did you discover the "ASI CRISP Control" plugin? It should make calibration of the CRISP a lot easier. Please do let me know how things work for you. I wrote the code last December and we used it successfully at the ASCB meeting with a spinning disk confocal and oil immersion objective.
